Search for: [Abstract = "The aim of these studies was to indicate how lipoic acid influences on the development of tolerance after longstanding nitroglycerin \(GTN\) medication in the rats and to try to explain the mechanism of the development of this tolerance to GTN in in vivo and in vitro studies. The studies contain\: an indication an influence of GTN and\/or lipoic acid on the rat’s blood pressure after one and longstanding medication, a creation tolerance to GTN in in vivo studies, an indication an influence of lipoic acid on the development of tolerance to GTN. A tolerance was induced by treatment of the rats with GTN at 10 mg\/kg, s.c., three times a day for 8 days. In animals treated jointly with GTN and lipoic acid at 50 mg\/kg, i.p., two times a day, for 8 days a tolerance didn’t occur. Probably a tolerance to GTN is because of the elevated reactive oxygen species production and breakdown of the defensive mechanisms of the organism. Therefore it was indicated an influence of the chronic GTN and\/or lipoic acid treatment on\: peroxidative processes and antioxidative enzymes, non\-protein thiols level, glutathione S\-transferase and γ\-glutamyl transpeptidase activities, NO and S\-nitrosothiols levels, sulfane sulfur level and rhodanese activity in the rats’ plasma and tissues. In the GTN\-tolerant rats the elevated reactive oxygen species and malondialdehyde levels and changed antioxidant enzymes activities in plasma and tissues were observed, whereas NO level in the aorta and plasma, Snitrosothiols level in plasma and glutathione S\-transferase activity in plasma and heart were decreased. Combined treatment with GTN and lipoic acid could prevent these adverse changes."]
