Evidence for a genetic basis for stroke isderived from studies of twins and relatives of people whohave had a stroke.The purpose of this thesis was to analyze the role ofselected polymorphisms of three candidate genes aspotential risk factors for stroke in the Polish population andto summarize the current knowledge on monogenic diseasesthat cause stroke.Polymorphisms of three selected genes were analyzed inthis study. Three common polymorphisms in plasminogenactivator inhibitor\-1 \(PAI\-1\) gene \(\-844 G.A, \-675 4G .5G,and HindIIIG.C\) were studied in patients with ischemicstroke due to large vessel disease, small vessel disease orcardioembolism. Glycoprotein IIIa \(GpIIIa\) gene A1\/A2polymorphism was studied in patients with hemorrhagicstroke caused by rupture of an aneurysm. Angiotensin IItype 1 receptor \(AGTR1\) gene A1166C polymorphism wasstudied in patients with hemorrhagic stroke, caused by thetwo most common types\: intracerebral hemorrhage andsubarachnoid hemorrhage.A\-G\-4G haplotype of PAI\-1 gene was found to beassociated with an increased risk of ischemic stroke in smallvessel disease in the Polish population. The GpIIIa A1\/A2polymorphism was not a risk factor of aneurysmalsubarachnoid hemorrhage in the Polish population. AGTR1A1166C gene polymorphism was not a risk factor forhemorrhagic stroke, while the AA genotype proved to beassociated with better outcome in aneurysmal subarachnoidhemorrhage in the Polish population."]
